      【目的】为了全面掌握乌鸦果组培快繁技术,促进我国乌鸦果野生种质资源保存和合理开发利用,对2个乌鸦果居群进行了组织培养和快速繁殖技术研究。 【方法】 以居群土壤pH值分别为5.90、7.68的2个乌鸦果居群幼嫩枝条茎段作为外植体,在pH值5.5且添加不同激素的WPM培养基上进行初代培养、继代培养及瓶内生根培养,采用不同生根剂及基质进行瓶外生根。【结果】实验表明,居群土壤pH值5.90和7.68的2种乌鸦果在pH值5.5的培养基中均能较好地诱导分化及增殖,只加玉米素而不加生长素更有利于乌鸦果诱导分化及增殖;但是2个乌鸦果居群瓶内生根培养差异较大,居群土壤pH值7.68的乌鸦果试管苗在pH值5.5的培养基中极难生根,居群土壤pH值5.90的乌鸦果试管苗在pH值5.5的培养基中能正常生根;2个乌鸦果居群瓶外生根时均能较好生根,两者差异较小。【结论】WPM+1.0 mg/L ZT是2个乌鸦果居群初代培养较佳的诱导培养基;低浓度ZT更有利于2个乌鸦果居群增殖,WPM+0.5 mg/L ZT是2个乌鸦果居群较佳的增殖培养基;不同IBA浓度对2个乌鸦果居群瓶内生根生根率影响差异较显著,1/2WPM+0.02 mg/L IBA是鲁甸乌鸦果居群较佳的生根培养基,居群土壤pH值7.68的贵峰居群乌鸦果在pH值5.5的生根培养基里极难生根;采用瓶外生根方法时,2个乌鸦果居群生根情况良好,但在同等条件下,乌鸦果瓶外生根时间比蓝莓栽培品种晚7 d以上,居群土壤pH值呈碱性的贵峰乌鸦果居群则比鲁甸乌鸦果居群更晚4 d左右;进口苔藓及IBA-K较有利于2个乌鸦果居群试管苗瓶外生根。

      【Objective】To comprehensively master the tissue culture and rapid propagation techniques of Vaccinium fragile Franch. and promote the conservation and sustainable utilization of its wild germplasm resources in China, a study was conducted on the tissue culture and micropropagation technology of two V. fragile Franch. Populations.【Method】Using young stem segments from two V. fragile Franch. populations with soil pH values of 5.90 and 7.68 as explants, primary culture, subculture, and in vitro rooting were conducted on WPM medium with pH 5.5 supplemented with different hormones. Additionally, different rooting agents and substrates were applied for ex vitro rooting. 【Results】The experiments demonstrated that both V. fragile Franch. populations originating from soils with pH 5.90 and 7.68 exhibited favorable differentiation and proliferation in the pH 5.5 culture medium. The medium supplemented with zeatin alone without auxin is more favorable for inducing differentiation and proliferation in V. fragile Franch.. However, significant differences were observed in their in vitro rooting performance. The test-tube seedlings from the population with soil pH 7.68 showed extremely poor rooting in the pH 5.5 medium, whereas those from the population with soil pH 5.90 rooted normally under the same conditions. In contrast, both populations exhibited successful ex vitro rooting with minimal differences between them. 【Conclusion】 The optimal induction medium for initial culture of both V. fragile Franch. populations were WPM supplemented with 1.0 mg/L ZT. A lower concentration of ZT was more conducive to the proliferation of the two populations, with WPM + 0.5 mg/L ZT being the optimal proliferation medium. The effect of different IBA concentrations on the in vitro rooting rate showed significant differences between the two V. fragile Franch populations.For the Ludian population of V. fragile Franch., 1/2 WPM + 0.02 mg/L IBA was the better rooting medium, while the Guifeng population with pH 7.68 showed extremely poor rooting in a rooting medium with pH 5.5. When using ex vitro rooting methods, both populations exhibited good rooting performance. However, under the same conditions, V. fragile Franch. required over 7 days longer for ex vitro rooting compared to cultivated blueberry varieties, and the Guifeng population took about 4 days longer than the Ludian population. Imported sphagnum moss and IBA-K were found to enhance ex vitro rooting in both V. fragile Franch. populations.
