| 摘要:糖类在果实生长发育、品质形成和营养积累过程中发挥关键调控作用。为了深入解析珍珠枣油桃糖代谢机制,本研究选取幼果期、膨大期和成熟期三个关键发育阶段的果肉组织,通过整合生理生化测定与转录组测序分析,系统检测了葡萄糖、果糖、蔗糖和山梨醇的含量动态变化,并深入挖掘了糖代谢相关基因的表达模式。结果表明:不同时期珍珠枣油桃果肉中葡萄糖、果糖和蔗糖成熟期含量高于幼果期和膨大期,而山梨醇的变化趋势相反;在珍珠枣油桃果肉中BvsA(膨大期vs幼果期)、CvsA(成熟期vs幼果期)、CvsB(成熟期vs膨大期)分别鉴定到8004、10114、6238个差异表达基因(DEGs)。KEGG富集分析参与糖代谢的差异基因主要包括1个蔗糖合成酶(SuSy) 、3个蔗糖转化酶(INV)、1个淀粉磷酸化酶(SP)、1个淀粉合成酶(SS)、2个腺苷二磷酸葡萄糖焦磷酸化酶 (AGPase)、3个果糖激酶(FRK)以及2个核酮糖-1,5-二磷酸羧化酶/加氧酶(Rubisco)。本研究阐明了珍珠枣油桃发育过程中糖积累与代谢基因表达的协同变化规律,为进一步解析珍珠枣油桃糖代谢调控网络提供了新的理论依据。 |

| Abstract: Carbohydrates play a crucial regulatory role in the growth and development, quality formation, and nutrient accumulation of fruits. To deeply analyze the sugar metabolism mechanism of pearl jujube nectarine, this study selected the flesh tissues of three key developmental stages-the juvenile fruit stage, the expansion stage, and the mature stage. Through integrating physiological and biochemical measurements with transcriptome sequencing analysis, the dynamic changes in the contents of glucose, fructose, sucrose, and sorbitol were systematically detected, and the expression patterns of sugar metabolism-related genes were deeply explored. The results showed that the contents of glucose, fructose, and sucrose in the flesh of pearl jujube nectarine were higher in the mature stage than in the juvenile fruit stage and the expansion stage, while the change trend of sorbitol was opposite. In the flesh of pearl jujube nectarine, 8004, 10114, and 6238 differentially expressed genes (DEGs) were identified for BvsA (expansion vs. juvenile fruit), CvsA (mature vs. juvenile fruit), and CvsB (mature vs. expansion) respectively. KEGG enrichment analysis showed that the differentially expressed genes involved in sugar metabolism mainly included 1 sucrose synthase (SuSy), 3 sucrose invertases (INV), 1 starch phosphorylase (SP), 1 starch synthase (SS), 2 adenosine diphosphate glucose pyrophosphorylase (AGPase), 3 fructose kinases (FRK), and 2 ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco). This study clarified the coordinated changes in sugar accumulation and metabolism gene expression during the development of pearl jujube nectarine, providing a new theoretical basis for further analyzing the sugar metabolism regulatory network of pearl jujube nectarine. |
