| 【目的】探究不同发育时期莲雾果实转录组中SSR位点信息和分布特征。【方法】对不同发育时期莲雾果实进行转录组测序,通过MISA软件鉴定莲雾果实转录组序列SSR位点,并对SSR位点的分布特征进行分析。【结果】不同发育期莲雾果实转录组测序共获得48392条Unigenes,其中13285条Unigenes上鉴定到SSR位点,SSR位点的发生频率为27.45%。对SSR 序列的长度进行统计,发现 SSR 位点长度范围为 12-80 bp,平均长度为 20.95bp,平均分布频率为23.28%。SSR重复基元类型以二、三核苷酸重复最多,分别占总二核苷酸至六核苷酸重复数量的53.79%和 37.20%,重复基元的重复次数集中在 6~9次,比例高低依次为六次、五次、七次和八次。SSR优势重复基元中AG/CT(48.50%)出现频率最高,其次为 CCG/CGG (12.87%)、AGG/CCT(8.11%)和AAG/CTT(7.13%)基元类型,分布频率最低的重复基元为AAAG/CTTT(占比0.59%),其次分别为AAGG/CCTT(占比0.62%)、AGCG/CGCT(占比0.62%)和AAC/GTT(占比0.67%)。【结论】不同发育期莲雾果实转录组中存在丰富多样的 SSR 位点,该研究结果为EST-SSR标记的开发和果实品质形成的分子遗传机制研究提供数据基础。 |

| 【Objective】The study aimed to explore the SSR locus and distribution characteristics of Syzygium Samarangense fruit transcriptome at its different developmental stages.【Method】Transcriptome sequencing was performed on different developmental stages fruit of Syzygium Samarangense. The SSR locus and distribution characteristics of the transcriptome sequencing was identified by MISA software.【Result】A total of 48,392 Unigenes were obtained from transcriptome sequencing, of which SSR sites were identified on 13,285 Unigenes, and the frequency of SSR sites was 27.45%. It was found that the length range of SSR loci was 12-84 bp with an average length of 20.95bp by statistically analyzing. Dinucleotide and trinucleotide repeats were the most numerous among the SSR motif types, which was about 53.79% and 37.20% respectively. The number of motif repeats was concentrated in 6-9 times, with the highest frequency was six, five, seven and eight in the order. AG/CT (48.50%) was the most frequently occurring repeat motif among the SSR dominant repeat motifs, followed by CCG/CGG(12.87%), AGG/CCT(8.11%) and AAG/CTT(7.13%) motif types. AAAG/CTTT(0.59%) was the lowest frequently occurring repeat motif among the SSR dominant repeat motifs, followed by AAGG/CCTT(0.62%), AGCG/CGCT(0.62%) and AAC/GTT(0.67%) motif types. 【Conclusion】The SSR locus of Syzygium Samarangense fruit at different developmental stages were abundant and diverse. The research results would provide reference basis for developing EST-SSR functional marker primers and the molecular biological mechanisms of fruit quality formation. |
