| 为明确云南新平沃柑黑斑病致病菌,从云南新平各地区共计采集240枚患沃柑黑斑病叶片进行分离、纯化,选取部分侵染能力强的菌株进行鉴定确定病原菌种类,从中选择侵染能力最强的1株病原菌进行致病性验证明确致病性。结果表明,所分离出86株病原菌中选取的5株通过ITS1/ITS4扩增结果均在500-750 bp之间,Blast比对结果建立系统进化树表明均与叶点霉属首都叶点霉菌(Phyllosticta capitalensis)聚于一族,致病性验证表明叶片感病症状与田间感病症状吻合且生长迅速,发病率高达70 %,发病指数为40.8 %;果实感病症状略有差异,菌株生长状态与培养皿中一致,果实发病率为30 %,发病指数为18.3 %。将感病材料再次进行分离纯化与鉴定,结果与第一次相同,表明云南新平沃柑黑斑病致病菌确为首都叶点霉菌(P. capitalensis) |

| In order to identify the causal organisms of black spot disease of Citrus sinensis in Xinping, Yunnan, a total of 240 leaves with black spot disease of Citrus sinensis were collected from various areas of Xinping, Yunnan for isolation and purification, and some strains with strong infestation ability were selected for identification to determine the species of the pathogenic bacteria, and one strain of the pathogen with the strongest infestation ability was selected for pathogenicity verification to identify the causal organisms. The results showed that five strains of 86 pathogenic bacteria were amplified by ITS1/ITS4 with results between 500-750 bp, and the phylogenetic tree established by Blast comparison showed that all of them were clustered in the same family with Phyllosticta capitalensis, and the verification of the pathogenicity showed that the symptoms of leaf susceptibility coincided with those of field susceptibility and the growth was rapid. Pathogenicity verification showed that the symptoms of leaf pathogenicity coincided with those of field pathogenicity and grew rapidly, with an incidence rate as high as 70% and a pathogenicity index as high as 40.8%; the symptoms of fruit pathogenicity differed slightly, and the growth state of the strains was the same as that of the Petri dishes, with a fruit incidence rate of 30% and a pathogenicity index of 18.3%. The susceptible materials were isolated and purified again, and the results were the same as the first one, which indicated that the causal agent of black spot disease of Xinping bergamot in Yunnan was P. capitalensis. |
