| 【目的】建立一种基于乳胶微球免疫层析技术快速定量检测柑橘中咪鲜胺的方法。【方法】通过乳胶微球标记咪鲜胺单克隆抗体,结合硝酸纤维素膜上包被的咪鲜胺-牛血清白蛋白(T线)和羊抗鼠抗体(C线),构建免疫层析试纸条,并匹配读数仪实现对样品中咪鲜胺含量的快速定量检测。【结果】该方法定量范围为0.50~3.00 mg/kg,检出限为0.265 mg/kg,定量限为0.503 mg/kg,加标回收率86.0%~115.6%,变异系数5.8%~13.4%;试纸条对咪鲜胺锰盐、咪鲜胺-脱氨基咪唑和咪鲜胺-脱咪唑甲酰胺的交叉反应率分别为99%、103%、52%,可同时检测咪鲜胺及其含有2,4,6-三氯苯酚部分的代谢物;与气相色谱法相比,空白样本符合率100%,阳性样本结果符合率为74.8%~127.5%(R2=0.985)。【结论】本研究开发的乳胶微球免疫层析方法操作简便、灵敏度高、环境适应性好,适用于柑橘中咪鲜胺残留的现场快速筛查,为食品安全监管提供了有效的技术支持。 |

| 【Objective】To establish a rapid and quantitative method for the detection of prochloraz in citrus using latex microsphere-based immunochromatographic technology.【Method】Monoclonal antibodies against prochloraz were labeled with latex microspheres, and immunochromatographic test strips were constructed by coating nitrocellulose membranes with prochloraz-bovine serum albumin (BSA) conjugates (test line, T-line) and goat anti-mouse antibodies (control line, C-line). A strip reader was employed to achieve rapid quantitative detection of prochloraz in samples.【Results】The method exhibited a quantitative range of 0.50~3.00 mg/kg, with a detection limit of 0.265 mg/kg and a quantitation limit of 0.503 mg/kg. The spiked recovery rates ranged from 86.0% to 115.6%, and the coefficients of variation (CV) were 5.8%~13.4%. The cross-reaction rates with prochloraz-manganese chloride complex, prochloraz-desimidazole-amino and prochloraz- desimidazole-formylamino were 99%, 103% and 52%, respectively, enabling simultaneous detection of prochloraz and its metabolites containing 2,4,6-trichlorophenol. Compared with gas chromatography (GC), the method showed 100% consistency for blank samples and a result agreement rate of 74.8%~127.5% for positive samples (R2=0.985).【Conclusion】The developed latex microsphere immunochromatographic method is simple, highly sensitive, and environmentally adaptable, making it suitable for on-site rapid screening of prochloraz residues in citrus. This approach provides effective technical support for food safety regulation. |
