| 为保护和利用富民枳种质资源,以富民枳的茎段为外植体,进行离体再生体系建立研究,并对所得组培苗与实生苗的光合功能进行比较分析。结果表明,适宜腋芽诱导的最佳培养基为MT+6-BA2.0mg/L +IAA0.2mg/L+CH200mg/L,诱导率达88.9%,且长势良好;适宜不定芽增殖的最佳培养基为WPM+6-BA0.5mg/L+IAA0.5mg/L+AC0.2g/L,增殖系数为8.7;适宜生根诱导的培养基为MT+6-BA1.0mg/L+IBA0.5mg/L+AC0.2g/L,生根率为73.33%,生根系数为5.85。富民枳实生苗的胞间CO2浓度低于组培苗,组培苗的蒸腾速率、净光合速率、气孔导度均高于实生苗,且叶绿素含量也显著高于实生苗。相关性分析显示,胞间CO2浓度与叶绿素含量无明显相关性,SPAD值与叶绿素含量间存在较高正相关。本研究所得富民枳组培苗在昆明地区的光合功能表现优于实生苗。 |

| In order to protect and utilize the germplasm resources of Poncirus polyandra., the stem segment of Poncirus polyandra. was used as explants to establish in vitro regeneration system, and the photosynthetic function of the tissue culture seedling and the seedling was compared and analyzed.The suitable medium for axillary buds was MT+6-BA2.0mg/L +IAA0.2mg/L+CH200mg/L,the induction rate was 88.9%,the axillary buds grew well.The suitable medium for adventitious bud proliferation was WPM+6-BA0.5mg/L+IAA0.5mg/L+AC0.2g/L, the proliferation coefficient was 8.7.The suitable medium for rooting induction was MT+6-BA1.0mg/L+IBA0.5mg/L+AC0.2g/L,the rooting rate was 73.33% and rooting coefficient was 5.85.The intercellular CO2 concentration of the seedling was lower than the tissue culture seedling, the transpiration rate, net photosynthetic rate and stomatal conductance of the tissue culture seedling were higher than seedling, and the chlorophyll content of the tissue culture seedling was significantly higher than seedling.Correlation analysis showed that there was no significant correlation between intercellular CO2 concentration and chlorophyll content, but there was a high positive correlation between SPAD and chlorophyll content.The photosynthetic function of Poncirus polyandra. tissue culture seedling was better than that of seedling in Kunming area. |
