      利用荧光SSR标记鉴定芒果杂交后代的真实性,为芒果种质创新和新品种选育提供参考依据。【方法】以‘金煌’和‘热农1号’、‘金煌’和‘爱文’、‘热农1号’和‘台农1号’杂交后代及亲本为试验材料,采用荧光SSR标记结合毛细管电泳技术筛选亲本间具有差异的SSR引物,在苗期对其杂交后代单株真实性进行鉴定。【结果】3个杂交组合分别筛选出8、6、4对亲本间有差异的SSR引物,用于杂交种真实性鉴定。‘金煌’和‘热农1号’杂交F1代的7个单株在7~8个位点上扩增出双亲的多态性条带,鉴定为真杂种;‘金煌’和‘爱文’杂交F1代的6个单株在5个位点上扩增出双亲的多态性条带,鉴定为真杂种;‘热农1号’和‘台农1号’杂交F1代的11个单株均可在4个位点上扩增出双亲的多态性条带,鉴定为真杂种。荧光SSR分子标记技术能快速准确鉴定芒果杂交后代的真实性,可作为分子标记辅助育种手段用于杂交苗的早期筛选。
英文摘要:
      【Objective】 The authenticity of mango hybrid progenies was identified using fluorescent SSR markers, which will provide reference for mango germplasm innovation and new variety breeding.【Method】 The hybrid progenies and parent of ‘Jinhuang’ and ‘Renong No. 1,’ ‘Jinhuang’ and ‘Irwin,’ as well as ‘Renong No. 1’ and ‘Tainong No. 1’ were used as experimental materials, SSR primers with polymorphism between parents were screened. Fluorescent SSR markers were used in combination with capillary electrophoresis technology to identify the authenticity of individual plants of three F1 hybrid populations.【Result s】 The results showed that eight, six, and four pairs of SSR primers with polymorphism between parents were selected to identify the authenticity of hybrids in the three populations. Seven individuals could amplify polymorphic bands of both parents at seven to eight loci in the F1 progenies of ‘Jinhuang’ and ‘Renong No.1’, which were identified as true hybrids. Six individuals could amplify polymorphic bands of both parents at five loci in the F1 progenies of ‘Jinhuang’ and ‘Irwin’, which were identified as true hybrids. Eleven individuals could amplify polymorphic bands of both parents at four loci in the F1 progenies of ‘Renong No.1’ and ‘Tainong No.1’, which were identified as true hybrids. 【Conclusion】Fluorescent SSR molecular marker technology can quickly and accurately identify the authenticity of mango hybrid progenies, and can be used as a marker-assisted breeding method for early screening of hybrid seedlings.
