| 【目的】为了建立‘秋月’梨(Pyrus pyrifolia Nakai)简单、高效的组培快繁体系,为‘秋月’梨工厂化育苗提供技术支持。【方法】本研究以‘秋月’梨带芽茎段为外植体,采用完全随机试验,对外植体消毒时间,增殖和生根培养基进行筛选。【结果】本研究发现,HgCl2(0.1%)消毒5 min消毒效果较好,发芽率达55.00%。‘秋月’梨适宜的初代培养基为MS+2.0 g/L PVA+30.0 g/L蔗糖+7.0 g/L琼脂+1.5 mg/L 6-BA+0.1 mg/L NAA+0.4 mg/L GA3。最佳增殖培养基为MS+2.0 g/L PVA+30.0 g/L蔗糖+7.0 g/L琼脂+1.0 mg/L 6-BA+0.1 mg/L IBA,增值系数为3.48,平均芽长,株高和茎粗分别为9.02 mm、24.53 mm和2.60 mm。最佳生根培养基为1/2 MS+25.0 g/L蔗糖+6.0 g/L琼脂+0.7 mg/L IBA+0.7 mg/L NAA,生根率达81.25%,平均根系数量为4.20根,平均根长和直径分别为5.28 cm和1.97 mm。活性炭对‘秋月’梨组培苗生根影响很大,0.3、0.5、0.7 g/L的活性炭均会抑制生根。生根后,组培苗在草炭土:蛭石=1∶1的基质中移栽成活率最高(89.89%)。【结论】本研究建立了‘秋月’梨组培快繁技术,筛选了适宜‘秋月’梨的增殖、生根及炼苗移栽条件。 |

| 【Objective】In order to establish a simple and efficient tissue culture and rapid propagation system for ‘Akizuki’ pear, and provide certain technical support for the industrial production of ‘Akizuki’ pear. 【Methods】In this study, the bud stem stem segments with buds of ‘Akizuki’ pear were used as explants, explant disinfection time, proliferation and rooting medium were screened by completely randomized trial. 【Results】The results showed that the explants immersed in 1% HgCl for 5 minutes,had the best disinfection effect, with a survival rate of 55.00%. The medium for the initial culture for ‘Akizuki’ pear was MS + 2.0 g/L PVA + 30.0 g/L sucrose + 7.0 g/L agar + 1.5 mg/L 6-BA + 0.1 mg/L NAA + 0.4 mg/L GA3. The best subculture medium was MS + 2.0 g/L PVA + 30.0 g/L sucrose + 7.0 g/L agar + 1.0 mg/L 6-BA + 0.1 mg/L IBA, The proliferation coefficient was 3.48, the average bud length, plant height and stem diameter were 9.02 mm, 24.53 mm and 2.60 mm, respectively.
