      为探究LcZAT12在荔枝果实发育成熟中的作用,以禾荔及禾荔晚熟芽变突变株MS1、MS2作为材料,克隆出LcZAT12全长序列,并对其进行生物信息学分析。通过实时荧光定量技术,分析LcZAT12在荔枝不同组织器官及在果实发育时期中果皮和种子中的表达模式。实验结果显示,LcZAT12全长429bp,编码142个氨基酸;利用在线网站对LcZAT12的蛋白性质进行预测显示,该蛋白位于细胞核,磷酸化以丝氨酸为主,无信号肽与跨膜结构域,蛋白质二级结构以α螺旋和不规则卷曲为主;氨基酸同源序列比对结果显示LcZAT蛋白含有两个高度保守的结构域;系统进化树分析发现,LcZAT12蛋白与糖槭和漾濞槭的同源蛋白亲缘关系最近;时空表达模式表明,LcZAT12蛋白可能在荔枝果实发育期中起着关键作用。本研究旨在为拓宽荔枝熟期,培育优质荔枝品种提供理论依据。

      To investigate the role of LcZAT12 in the development and maturation of Litchi fruits, the full-length sequence of LcZAT12 was cloned from Heli and its late maturing bud mutant strains MS1 and MS2, and bioinformatics analysis was performed on them. By using real-time fluorescence quantitative technology, analyze the expression patterns of LcZAT12 in different tissues and organs of Litchi, as well as in the peel and seeds during fruit development. The experimental results show that LcZAT12 has a total length of 429bp and encodes 142 amino acids; Using online websites to predict the protein properties of LcZAT12, it was found that the protein is located in the nucleus, mainly phosphorylated with serine, without signal peptides or transmembrane domains. The secondary structure of the protein is mainly Alpha helix and Random coil; The amino acid homologous sequence alignment results showed that LcZAT protein contains two highly conserved domains; The phylogenetic tree analysis revealed that the LcZAT12 protein has the closest homologous protein relationship with the sugar maple and Yangbi maple; The spatiotemporal expression pattern suggests that the LcZAT12 protein may play a crucial role in the development of Litchi fruits. The aim of this study is to provide theoretical basis for expanding the ripening period of Litchi and cultivating high-quality Litchi varieties.
