| 目的 为 解析火龙果Hylocereus undatus 叶绿体基因组结构和序列特征,并探讨其系统进化关系。方法 ,运用高通量测序技术开展火龙果基因组测序,使用生物学信息技术对其进行组装和注释,分析其密码子偏好性、简单重复序列、与近缘物种间序列变异的关系,并构建系统进化树。 结果 结果表明,火龙果叶绿体基因组大小为133 071 bp,呈环状双链四段式结构,包括大单拷贝区68 001 bp,小单拷贝区21 716 bp 和两个12477 bp的反向重复区。火龙果叶绿体基因组共注释到 118 个基因,其中蛋白编码基因 76 个,rRNA基因 4 个、tRNA基因 38个。火龙果叶绿体基因组密码子偏好以 A/U(T)为结尾,在检测到的 66 个简单重复序列中,大多数是 A/T 碱基形成的单核苷酸重复基序。序列对比和系统进化分析显示火龙果与同科的硫磺团扇和长枪团扇具有较近的亲缘关系。结论 本研究首次测得了火龙果叶绿体基因组全序列,明确了其叶绿体基因组结构和序列特征,为火龙果的种植栽培和资源开发等研究提供了基因组信息。 |

| 【Objective】To illustrate structure and sequence characteristics of Hylocereus undatus chloroplast genome and explore its phylogenetic relationship. 【Method】The chloroplast genome of Hylocereus undatus was sequenced by high-throughput sequencing technology. Utilization of bioinformatics methods, the chloroplast genome sequence was assembled and annotated, and then codon preference, simple sequence repeat, and sequence variation between related species were analyzed. a phylogenetic tree was also constructed.【Result】The full size of the chloroplast genome was 133 071 bp, which is a circular double-stranded four-segment structure, including 68 001 bp of large single copy region, 21 716 bp of small single copy region, and two reverse repeat regions of 12477 bp. A total of 118 genes were annotated in the chloroplast genome, including 76 protein-coding genes, 4 rRNA genes and 38 tRNA genes. The chloroplast genome intended to used codon ended with A/U(T), and among 66 simple sequence repeat detected in the plastome, most were mononucleotide formed by A or T bases. Sequence comparison and phylogenetic analysis showed that Hylocereus undatus had a close relationship with Opuntia sulphurea and Opuntia quimilo.【Conclusion】The chloroplast genome of Hylocereus undatus were clarified for the first time, which would provide genome data for culture, breeding and resource development of this edible plant. |
