| 为了提高柑橘离体组织培养工作效率,利用特美汀(Timentin)等抑菌剂对成年态柑橘枝条外植体无菌化处理效果进行比较。结果表明,Timentin处理浓度≤500 mg/L时,对柑橘成年态枝条外植体的腋芽生长影响不大;浓度2%的NaClO处理后,外植体在培养过程中污染率变化不大;75%乙醇处理15 s+2% NaClO处理10 min+300 mg/L Timentin的处理条件下,冰糖橙外植体培养14天时的污染率为14.4%,无菌化培养出的柑橘外植体腋芽茎尖微芽嫁接后成活率能达到35.6%以上。本研究建立的柑橘成年态枝条无菌化培养方法污染率低,外植体腋芽活力高,能为进一步通过外植体组织培养加快柑橘良种脱毒等工作提供基础。 |

| In order to improve the efficiency of in vitro tissue culture for citrus, antibacterial agents such as Timentin were used for the bacteriostatic culture of adult citrus explants, and it was found that Timentin treatment concentration ≤500 mg/L had little negative impact on the axillary bud growth of the stems. After 2% NaClO treatment, the contamination rate of the stems did not change much during culture. Under the conditions of 75% ethanol treatment for 30 s+2% NaClO treatment for 20 min+300 mg/L Timentin, the bacterial contamination rate of the Bingtang orange explants cultured by 14 days was only 14.4%. The survival rate of the shoot-tip micro-grafting with buds cultured from explants was more than 35.6%. The method established in this study has low contamination rate and high axillary bud activity for adult citrus stems, which can provide a basis to accelerate the virus-free citrus varieties. |
