| 本研究以南高丛蓝莓品种戴安娜(Diana)组培苗为试材,设置0(CK)、20、50、100、150 μM不同浓度一氧化氮(nitric oxide, NO)供体硝普钠(sodium nitroprusside,SNP)处理蓝莓的茎段,分析培养40 d茎段基部愈伤组织的质膜透性、活性氧代谢、抗氧化产物含量和抗氧化酶活性等变化,培养70 d测定株高、茎鲜重、茎干重和叶绿素含量,计算分化出芽率和增殖倍数,以期探究外源NO供体SNP对南高丛蓝莓离体再生及理化特性的影响。结果表明:NO供体SNP对蓝莓瓶内苗的不定芽再生、增殖和生长有较明显的影响,但存在显著的浓度效应,20 μM SNP处理的始分化时间最短,分化出芽率、增殖倍数和叶绿素含量最高;50 μM SNP处理的鲜重和干重最高,150 μM SNP处理的株高、叶面积最大;同时20 μM SNP处理显著提高了茎段基部愈伤组织中SOD、GPX的活性,增加了非酶抗氧化物ASA和GSH含量,降低了O2-.产生速率和H2O2的积累,有效缓解了膜脂过氧化作用,减轻了茎基部愈伤组织的褐化,从而促进了戴安娜茎段的分化和增殖。综上所述,在WPM+ZT 2.0 mg/L培养基中添加20 μM SNP是戴安娜蓝莓不定芽再生和增殖的最佳处理。 |

| In this study, the tissue cultured seedlings of the southern blueberry variety Diana was used as the experimental material, and different concentrations(0, 20, 50, 100, 150 μM) of NO donor SNP were treated with the stem segments, and analyze the changes in plasma membrane permeability, reactive oxygen species metabolism, antioxidant product content, and antioxidant enzyme activity of callus tissue cultured at the base of stem segments for 40 days. and to measure plant height, fresh stem weight, dry stem weight, and chlorophyll content after 70 days of cultivation, and calculate the differentiation rate and proliferation multiple, in order to investigate the effects of exogenous nitric oxide donor sodium nitroprusside on in vitro regeneration and physicochemical properties from southern highbush blueberries. The results showed that exogenously supplied NO donor, sodium nitroprusside had significant positive effects on shoot differentiation, regeneration and proliferation
from double-node segments of Diana, excised from its in vitro seedlings, and the effect was dose-dependent with amaximal response. In particular, addition of 20 μM sodium nitroprusside gave shortest initial differentiation time, greatest shoot multiplication percentage and accumulation the highest contents of leaf chlorophyll, while 50 μM SNP exhibited the greatest shoot length and fresh and dry weight. However, 150μ M SNP caused the highest shoot length and leaf area. SNP applied at 20 μM led to raise superoxide dismutase and guaiacol peroxidase activities, the content of ascorbic acid and reduced glutathion. Whereas, the content of H2O2, production rate of O2–. and MDA level were decreased in callus at base of stem segment, and completely inhibiting the hyperhydricity symptoms of explants. Taking simultaneously into consideration all shoot proliferation parameters evaluated, the explants performance was optimum and significantly better among others when treated with WPM+2.0 mg/L ZT + 20 μM SNP. |
