      木奶果育苗过程中白绢病频发,严重影响木奶果种苗繁育。为明确病原菌种类、生物学特性和防治药剂,开展了本研究。首先,采用组织分离培养法分离病原菌,获得4株形态特征相似的菌株MNG-1~MNG-4,选取代表性菌株MNG-1进行致病性试验,结果符合柯赫氏法则。经形态学分析和病原菌DNA经引物ITS、TEF、SSU与LSU扩增的对比分析,鉴定病原菌MNG-1为罗尔阿太菌(Athelia rolfsii)。其次,对MNG-1的生物学特性进行分析,结果表明菌丝生长最适温度为30 ℃,最适pH值为7,最适培养基为PSA;菌核产生最适条件为低温、碱性和持续光照。最后,以菌丝生长速率法进行防治药剂筛选,结果表明噻呋酰胺对MNG-1菌丝的抑制效果最好,EC50为0.036 mg·L-1,其次是戊唑醇,EC50为0.463 mg·L-1,恶霉灵、甲基硫菌灵、百菌清、代森锰等杀菌剂对该菌的抑制效果稍差。
英文摘要:
      Southern blight of Baccaurea ramiflora occured frequently in the seedling raising process, which seriously affected the seedling propagation of Baccaurea ramiflora. This study aims to identify the pathogen, clarify the biological characteristics, and screen the effective fungicides. Above all, the tissue culture method was applied to isolate the pathogen and the results showed the four strains MNG1~MNG4 were obtained based on the similar morphological features, a representative MNG-1 strain was selected for the pathogenicity test and fulfilled the Koch’s postulates. The pathogen MNG-1 was identified as Athelia rofsii by the morphology and Comparative analysis of DNA amplification of pathogenic bacteria by primers ITS, TEF, SSU and LSU. Secondly, the biological characteristics were analyzed that the optimum growth temperature for MNG-1 was 30 ℃ and pH was 7, the optimal medium was PSA; the optimum conditions for the generation of sclerotium are low temperature, alkalinity and continuous light. At last, mycelium growth rate method was used to determine the most effective fungicides. The results of laboratory virulence test showed Thiafuramide has the best inhibitory effect on MGN-1 mycelium, that the EC50 was 0.036 mg·L-1. next is Tebuconazole, that the EC50 was 0.463 mg·L-1. The inhibitory effect of southern blight disease was slightly worse, such as Hymexazol, Thiophonate-methy, Chlorothalonil and Mancozeb.
