| 本试验以中柑所5号及其亲本为材料,采用SSR分子标记技术,筛选出适用于中柑所5号鉴定的核心引物9对,其中5对引物(CSS038、SS16、CSS052、SS9、CSS014)表现同父本带型,3对引物(CSS020、SS12、CSS037)则表现同母本带型,1对引物SS2出现父母本之外的新带型,建立了基于SSR分子标记的中柑所5号的DNA指纹图谱。利用筛选出的引物和DNA指纹图谱,我们鉴定出两个分别来自资阳和简阳的某苗圃的未知苗木样本为中柑所5号。 |

| In this study, 9 primer pairs for Simple Sequence Repeat (SSR) molecular markers were first screened out and then used to construct the fingerprinting of CRIC 5 (Citrus Reticulata Blanco Cv. Ehime No. 30 × Citrus Reticulata Blanco Cv. Shiyueju). In these screened primer pairs, 5 primer pairs (CSS038、SS16、CSS052、SS9、CSS014) showed the same bands with its male parent Shatangju, 3 primer pairs (CSS020, SS12, CSS037) with its female parent Ehime No. 30, and 1 primer pair is different from both male and female parents. By using these primers and fingerprinting, two unknown citrus samples (from Jianyang and Ziyang, Sichuan, respectively) was identified as citrus CRIC 5. Thus, DNA molecular marker technique based on SSR is a powerful tool for protecting this new citrus cultivar. |
